SKRIPSI KARAKTERISASI ISOLAT BV-3PENGHASIL SIKLODEKSTRIN GLUKANOTRANSFERASE""
ABSTRAKrnrnEnzim siklodektrin glukanotransferase (CGTase) adalah enzim ekstraseluler yang diproduksi oleh beberapa jenis bakteri dan archae.CGTase mengubah pati menjadi siklodektrin (CD) yang berperan penting dalam bidang farmasi.Peneliti sebelumnya telah berhasil menemukan isolat bakteri dari tanah perkebunan singkong di Jambi yang disebut isolat BV3.Penelitian ini bertujuan untuk mengetahui karakter lama inkubasi, suhu inkubasi, dan pH media pertumbuhan isolat BV-3 dalam produksi β-CGTase dan karakter crude enzyme-nya. Optimasi pertumbuhan isolat BV-3 dalam produksi β-CGTase dilakukan pada lama inkubasi 24, 48, dan 72 jam; suhu inkubasi 30oC, 37oC, 50oC, dan 60oC; serta pH media pertumbuhan pH 6, 7, 8, 9, 10, dan 11. Hasil penelitian menunjukkan bahwa isolat BV-3 menghasilkan β-CD yang optimum jika diinkubasi selama 48 jam, suhu inkubasi 30oC, dan pH media pertumbuhan 10. Crude enzymeβ-CGTase dari isolat BV-3 juga dikarakterisasi terhadap suhu dan pH dalam rentang 30-60oC dan pH 6-10.Crude enzymeβ-CGTase dari isolat BV-3 memberikan aktifitas yang besar yaitu sebesar 302.76 µg/mL pada suhu 60oC pH 8.rnKata Kunci : Siklodekstrin glukanotransferase (CGTase), Crude Enzyme, Optimasi Pertumbuhan, Optimasi EnzimrnABSTRACTrnThe cyclodextrin glucanoltransferase (CGTase) is an extracellular enzyme which is produced by several types of bacteria and archae. CGTase converts starch into cyclodextrin (CD), which plays an important role in the pharmaceutical industries. The previous researcher have found a CGTase bacteria from cassava soil, named BV-3 isolate. The aim of this research was to find out the character of incubation period, incubation temperature, and pH of BV-3 isolate growth medium for β-CGTase production and the character of crude enzyme β-CGTase based on pH and temperature parameter.The optimization of BV-3 growth for β-CGTase production was performed at 24, 48 and 72 hours incubation time; Incubation temperature was performed at 30oC, 37oC, 50oC, and 60oC; And pH of growth media was performed at pH 6, 7, 8, 9, 10, and 11. The result of growth optimization showed that BV-3 produce the largest amount of β-CD if incubated for 48 hours, at temperature 30oC, and pH 10. The crude enzyme β-CGTase produced by BV-3 was also characterized by temperature and pH parameters in the ranges of 30-60oC and 6-10, respectively.
